The effects of pentoxifylline, pentifylline and gamma-interferon on myofibroblast proliferation among other things.
Myofibroblasts are important for wound healing, these cells die at the end of woundhealing, but in Peyronies Disease (and Dupruytens) they don't die etc..

In this study pentifylline was much much more potent than pentoxifylline.

What I am learning from the link emasculated is that you need 10 times higher dose of Pentoxifylline than Pentifyline.
So maybe instead of let say 3*400 mg Pentox per day, one should take 3*40 mg Pentifylline.
Interesting how will be the Pentifylline side effects and if it also suppressing the immune system.
I stopped Pentox because I was getting flue every two months and more and more bad.

james: Cosaldon comes in the same dosage as Trental. 3x400 mg daily and has the same possible side effects. It's just much more effective. That's my reading of it. Someone correct me if I made a mistake here.

I see that pentifylline is 10 times stronger than Pentox when it comes to fibroblast but does it have the same ability as Pentox to effect the transport of blood cells into the smallest blood vessels?

@MattFoley: Only a study can decide that. But if in doubt I don't think combining Pentox with Cosaldon would be a problem. If one takes say 3x800 mg Pentox daily anyway, one could take instead 3x400 mg Pentox combined 3x400 mg Cosaldon daily. Any volunteers? ;-)
Of course the study I posted is about fibrosis in the kidney and we don't know how much still applies to our problem.. yet still.. there should be a study comparing Placebo, PTX and PTF in Peyronies Disease.

EDIT: I guess since funds are so limited we will never find out with a study whether this medication is more effective with Peyronies Disease. :-( I would still be VERY interested in someone's opinion who is more knowledgable than me.

James, my general practitioner told me that he was a researcher on Pentox before becoming a doctor. He told me that if I get an infection, I should let him know right away. Pentox somehow affects our ability to fight infections. This might be why you get the flue when using it?

Yes IhatePD, the immune system is affected by Pentox, this is the reason why after 18 months (by the way with a lot of benefits I detailed a few time already on the forum) stopped Pentox. After stopping three months ago, no flue thanks GOD.

I've noticed a few people complain that Pentox makes them a bit fuzzy mentally. Here's an old article (1982) that suggests Pentifylline has the opposite effect:

Abstract


The effect of a single dose of 4 sugar-coated tablets of Cosaldon (1-hexyl-3,7-dimethylxanthine (pentifylline) + nicotinic acid) was shown by quantitative pharmaco-EEG investigations and psychometric tests in 12 volunteers aged 58 to 75 years, who were in a good physical condition. The design was randomized, double-blind and cross-over designed against placebo. EEG and psychometric tests were made before medication and repeated each hour up to 6 h after medication. Each turn was split up into a resting-EEG (R-EEG) and a vigilance-EEG (V-EEG) lasting 3 min each; the psychometric tests were made immediately afterwards. An increase of the vigilance, shown by an increased EEG-power, was found after Cosaldon administration as compared to placebo. Furthermore, the delta and theta intensity decreased to a statistically significant degree, while the intensity within the alpha-1-scope increased. The strongest effect as compared to placebo could be observed 4 h after medication. The statistical significance was shown for the V-EEG-condition, the R-EEG showed only minor changes. The effects in the occipital region were stronger than in the more frontal parts. In accordance with the results of the EEG, the psychological tests showed remarkable improvements in performance which reached their maximum between 2 and 4 h after medication.
